Investigation of Anticonvulsant Effects of Some Medicinal plant Hydrosols

Epilepsy is one of the most common chronic neurological disorder that affects people of all age groups. A number of medicinal plants are used in traditional European and Iranian folk medicines to treat some of neurological diseases including epilepsy. For this study, Dried clove flower buds, valeria roots, cinnamon barks, yarrow areal parts, black seed fruits, galbanum roots and golpar seeds were subjected to hydrodistillation and hydrosols were extracted by a solvent in a separating funnel to obtain the organic phase of hydrosol (OPH). Finally, OPH content and compositions were investigated with gas chromatography techniques. The OPH of all samples were tested for potential anticonvulsant activity using the PTZ epilepsy zebrafish model. The results showed the clove OPH revealed the strongest anticonvulsant activity, decreasing PTZ-induced locomotor activity at ۳ μg/mL tested (p<۰.۰۰۱). The valerian and golpar OPH also decreased locomotor activity at ۱۰۰ μg/mL (p<۰.۰۰۱ for valerian OPH and p<۰.۰۱ for golpar OPH), while at ۲۰۰ μg/mL it induced toxicity with an augmentation of fish movements. In contrast, the galbanum (۱۰۰ μg/mL), black seed (۱۰۰ μg/mL) and valerian (۳۰ μg/mL) were not active and valerian OPH exhibited a significant increase in locomotor activity at the ۳۰ μg/mL concentration tested. In fact, results shows that anticonvulsant properties of the active hydrosols could be attributed to the main volatile compounds like eugenol, anethole, pulegone, borneol and limonene.
